The use of cellular models in the prediction of the role of rowanberry phytonutrients in the regulation of female reproductive functions

Abstract
Due to the influence of the environmental burden and risk factors of the food chain, the risk of developing reproductive diseases increases. The most common diseases affecting the female reproductive system include infertility, fibroids, cervical carcinomas, ovarian tumors, endometriosis, endocrine disorders, polycystic ovary syndrome and premature ovarian failure with a negative impact on quality of life. Natural substances present in plants, spices, fruits can effectively modulate key elements of the signaling pathways of viability, steroidogenesis, proliferation and apoptosis in the female reproductive system and eliminate negative environmental factors. The targeted combination of phytosubstances verified using cell models (human ovarian cell lines, cervix) are promising sources for supporting hormonal regulation, improving reproductive functions, as well as preventing reproductive alterations in females.
The scientific focus of the proposed project is divided into 3 stages:
Stage 1 will be focused on experimental testing of the in vitro effects of rowanberry phytonutrients (Sorbus aucuparia / Sorbus domestica L.) on model cells of the female reproductive system (ovarian granulosa cell line HGL5, ovarian tumor cells OVCAR-3 and COV434, cervical tumor cells HeLa and i.) for determining viability and secretory activity (growth factors, steroid hormones and i.).
Stage 2 , based on the results of Stage 1, will be aimed at determining the mechanisms of action of the rowanberry phytonutrients (Sorbus aucuparia / Sorbus domestica L.) on cell models of the female reproductive system through signaling pathways interfering with the processes of proliferation and apoptosis (receptors, markers of proliferation and apoptosis).
Stage 3 will be focused on the determination and selection of experimentally tested phytonutrients and their combinations with high cytotoxic activity on tumor cells with the aim of inducing programmed cell death - apoptosis and elimination of cell growth, proliferation and metabolism without negative impact on healthy cells.
Scientific hypothesis: The targeted combination of rowanberry phytonutrients verified by the use of cell models are promising sources for the support of hormonal regulation, improvement of reproductive functions, as well as prevention of reproductive alterations in females.
Využitie bunkových modelov v predikcii úlohy fytonutrientov plodov jarabiny v regulácii reprodukčných funkcií samíc
Abstrakt
Vplyvom environmentálnej záťaže a rizikových faktorov potravového reťazca narastá riziko vzniku reprodukčných ochorení. Najčastejšie ochorenia postihujúce samičí reprodukčný systém zahŕňajú neplodnosť, myómy, karcinómy krčka maternice, nádory vaječníkov, endometriózy, endokrinné poruchy, syndróm polycystických vaječníkov a predčasné zlyhanie vaječníkov s negatívnym dopadom na kvalitu života. Prírodné látky prítomné v rastlinách, koreninách, ovocí dokážu efektívne modulovať kľúčové prvky signálnych dráh viability, steroidogenézy, proliferácie a apoptózy v samičom reprodukčnom systéme a eliminovať negatívne faktory prostredia. Cielená kombinácia fytosubstancií verifikovaných využitím bunkových modelov (humánne bunkové línie vaječníkov, krčka maternice), sú perspektívne zdroje pre podporu hormonálnej regulácie, zlepšenie reprodukčných funkcií, ako aj prevenciu reprodukčných alterácií samíc.
Vedecké zameranie navrhnutého projektu je rozdelené do 3 etáp:
Etapa 1 bude zameraná na experimentálne testovanie in vitro účinkov prírodných látok plodov jarabiny (Sorbus aucuparia / Sorbus domestica L.) na modelových bunkách reprodukčného systému samíc (ovariálna línia granulóznych buniek HGL5, ovariálne nádorové bunky OVCAR-3 a COV434, nádorové bunky krčka maternice HeLa a i.) pre stanovenie viability a sekrečnej aktivity (rastové faktory, steroidné hormóny a i.).
Etapa 2 , na základe výsledkov Etapy 1, bude zameraná na určenie mechanizmov účinku fytonutrientov plodov jarabiny (jarabina vtáčia / jarabina oskorušová) na bunkové modely reprodukčného systému samíc prostredníctvom signálnych dráh zasahujúcich do procesov proliferácie a apoptózy (receptory, markery proliferácie a apoptózy).
Etapa 3 bude zameraná na určenie a selekciu experimentálne testovaných fytonutrientov a ich kombinácií s vysokou cytotoxickou aktivitou na nádorové bunky s cieľom indukcie programovej bunkovej smrti – apoptózy a eliminácie bunkového rastu, proliferácie a metabolizmu bez negatívneho vplyvu na zdravé bunky.
Vedecká hypotéza: Cielená kombinácia fytosubstancií plodov jarabiny verifikovaných využitím bunkových modelov sú perspektívne zdroje pre podporu hormonálnej regulácie, zlepšenie reprodukčných funkcií, ako aj prevenciu reprodukčných alterácií samíc.
