The Research Unit Translational Immunology (TUM) in collaboration with the Institute of Lung Health and Immunity (LHI; Helmholtz Munich) is looking for a senior scientist / postdoc (PhD) (m/f/d) to study pulmonary delivery of gene editing tools and successful genetic and functional correction of STAT3 mutations in lung cancer cells and iPSC-derived alveolar epithelial cells.

The Research Unit Translational Immunology (TUM) and the Institute of Lung Health and Immunity (LHI; Helmholtz Munich) joined forces to better understand and cure lung diseases associated with a dysregulated STAT3 signaling using CRISPR-mediated gene editing. The proposed project focuses on STAT3 gene correction in lung cancer and STAT3-hyper-IgE syndrome (STAT3-HIES), which is a monogenic inborn error of immunity with chronic destructive lung disease. To correct the underlying disease-causing STAT3 mutations we established CRISPR/Cas9-mediated base editing (Eberherr et al. CRISPRJ).

Your responsibilities:

  • Establish air-liquid interface (ALI) culture of iPSC-derived alveolar epithelial cells
  • Perform gene editing of human lung cancer cells and iPSC-derived alveolar epithelial cells using polymers developed at the lab of Prof. Merkel (Drug Delivery, LMU)
  • Assess efficiency and safety of the delivery method and the performed CRISPR-mediated gene editing

Your profile:

  • PhD in one of the life sciences or equivalent work experience in the wet lab
  • A strong background in polymer-based cellular delivery of nucleic acids or proteins and cell culture technique is favorable
  • Ideally you have experience in gene editing (including gRNA design and validation), ALI and/or iPSC culture
  • Experience in downstream analysis including immunofluorescence staining, flow cytometry, western blot analysis, quantitative real-time PCR, Sanger sequencing, high-throughput sequencing, in silico analysis of editing efficiency, cell viability analysis is a plus
  • The highly motivated candidate must be able to work independently however we greatly seek team player mentality
