Projects: Studying the nanoscale organization of B lymphocyte surface receptors and neuronal synapses using nanobodies and super-resolution imaging approaches
One project's primary goal is to achieve a comprehensive picture of the spatial organization of all BCR isotypes in conjunction with their essential coreceptors and signaling-active lipids within the plasma membrane at nanoscale resolution. The second project aims to characterize the nanoscale of neuronal synapses using self- generated nanobodies revealing various synaptic targets.
Both projects require the discovery process of specific nanobodies recognizing endogenous proteins and use their monovalency to obtain quantitative STED, DNA- PAINT, and Expansion microscopy (ExM). Ideally, we also want to combine these super-resolution techniques with nanoSIMS. to monitor the surrounding lipids of transmembrane protein (nanoSIMS is a mass-spectrometry imaging technique with ~50 nm precision in XY and ~5-10 nm in Z)
What we are looking for:
We are looking for highly motivated candidates with broad interests, from developing and characterizing new affinity molecules like nanobodies (skills: molecular biology and protein biochemistry) to learning and using several microscopy techniques (DNA- PAINT, STED, ExM, and nanoSIMS) for imaging immune cells (B lymphocytes) and or cultured neurons.
