This thesis will be carried out at the IAB, Grenoble in the group headed by Olivier DESTAING within the DYSAD team at the IAB in conjunction with its 3 tutelles (CNRS, UGA, Inserm). This work will be part of a national collaboration with Dr. C. Brun of TAGC-Marseille, Dr. D. Bourgeois of IBS-Grenoble and prof. Fourcade of Liphy-Grenoble, and will be open to international collaborations (USA and Canada). The student will benefit from our collaboration with the EDyP proteomics facility at CEA Grenoble and the microscopy facility at IAB (MicroCell).

The aim of this PhD project is to gain a better understanding of the molecular basis of SRC signaling by combining optogenetic activation of our optoSFKs with proximity labeling (Apex, biOID) and phosphoproteomics. Our laboratory is expert in the optogenetic control of cell signaling and has developed numerous optogenetic probes to control each member of the SFK. The proximity labeling approach (Apex, biOID) is a fast-developing technique. It enables the close molecular environment of a protein of interest to be identified directly in the living cell, thanks to its labeling by biotinylation. The tagged proteins are then extracted and analyzed by mass spectrometry. We will couple our optoprobes to both proximity labeling analysis and phosphoproteomics. This will enable us to specifically activate a signaling event (downstream activation of a specific optokinase) and then map both the molecular environment associated with the optoprobe and its information transfer by identifying downstream phosphorylated proteins. The data obtained will then be analyzed using a dedicated network of interactomic networks developed jointly with Dr. C. Brun of TAGC-Marseille.
