DNA damage induced by Sunlight and tumor transformation.

The Institute of Biomedical Sciences, University of São Paulo, has a postdoctoral position, with a scholarship from FAPESP, under the Thematic Project "Role of DNA damage and mitochondrial function in vascular, immune and neurological aging." (DNA MoVINg - process 2019/19435-3). This project will be developed under the supervision of Dr. Carlos Frederico Martins Menck and applications will be received until June 30th , 2021.

The project to be developed intends to investigate tumor transformation mechanisms in cells deficient in DNA repair or translesion synthesis processes, notably xeroderma pigmentosum (XP), due to DNA damaged induced by sunlight. As a study model, cells from affected patients will be evaluated, after irradiation with light of different wavelengths, to search for events that may result in tumor transformation. We are particularly interested in mechanisms of oxidation of DNA repair proteins, with inhibition of damage removal. We are predicting studies that will allow the identification of possible novel mechanisms of transformation and phenotypes involved. Subsequently, the candidate will assess mutagenesis levels through new generation sequencing (NGS). Thus, we will seek to correlate DNA lesions and their effects on the generation of tumor phenotype in these patients.

Candidates must have previous experience in human cell culture, and preferentially capacity to evaluate, select and quantify tumor phenotypes. Must also know cell and molecular biology; previous work on DNA repair processes and in methodologies for detecting DNA damage, cell and mitochondrial metabolism, and assessing the formation of oxygen radicals (ROS) in cells will also be considered. The candidate must develop research involving techniques of cell biology, molecular biology, protein, and DNA extraction, and preparation of DNA library for new generation sequencing, in vitro assays, and cell culture. As for training, candidates with PhD in Genetics, Molecular Biology, Biochemistry or related area are required for the vacancy.

The opportunity is open to Brazilians and foreigners. The selected candidate will receive a Postdoctoral Scholarship from FAPESP, for a period of 02 years (renewable for up to 4 years) in the amount of R $ 7,373.10 per month and overhead, which is equivalent to 15% of the annual value of the scholarship and has the objective to meet unforeseen expenses and directly related to the research activity.
