DFCI has pioneered the first generation of personal cancer vaccine, which have shown great promise, and our mission is now to develop a second-generation program which overcomes key hurdles (time and cost of manufacturing) and improves efficacy (including a broader repertoire of targeted neoantigens).
We are seeking a highly skilled and motivated scientis to develop and conduct the molecular biology experiments critical to our next generation personal cancer vaccine program.?? The novel approach we envision relies extensively on both conventional and cutting-edge nucleic acid molecular biology techniques to prepare and evaluate the vaccine. The?? applicant will work closely with immunologists and cell biologists to design and prepare molecules for testing in vitro and in animal models, culminating in translation to a human clinical trial.

- The individual will be critically involved in all aspects of the immunology and molecular biology of the program, ranging from extraction of RNA from cell lines and patient samples, preparation of cDNA libraries, the design and construction of large and diverse plasmid libraries, gene expression in mammalian cells and applying a variety of immunological techniques to analyze immunogenicity and immune cell activation and phenotype.
- Utilize their existing knowledge, search the literature and query relevant colleagues to identify improvements to the currently envisioned steps as well as solve emerging problems.
- Interact technically and intellectually with bioinformatic scientists to produce and analyze nucleic acid sequencing data to determine the diversity and complexity of produced libraries relative to expectations
- Collaborate with immunologists in the group to design and produce materials to be used in cell culture and animal studies
- Work with internal and external development and manufacturing groups to design and test the molecular biology process steps and analytics to produce a vaccine for a human clinical trial.
- MS or PhD with experience in an academic or biopharmaceutical laboratory applying a variety of immunology (flow cytometry, vaccination, immune cell isolation and analysis) and molecular biology techniques, (PCR, plasmid subcloning, multi-step protocols involving various enzymatic reactions and separation steps). Demonstrated knowledge of the principles involved for each step. Some experience with protein biochemistry preferred.
- Understanding and experience with generating, organizing and analyzing sequencing data.
- Capable of understanding broad goals and defining the experiments needed to meet those goals. Ability to independently organize, effectively conduct, critically interpret and fully document experimental plans without day to day direct involvement of a supervisor.?? Exercises good judgement when additional guidance is required.
- Ability to effectively communicate both 1:1 with supervisor and other team members as well as prepare and deliver clear and critical summaries of their work.
- Works collaboratively as part of a team.
Demonstrates potential for technical proficiency, scientific creativity, collaboration with others and independent thought. Works on problems of moderate to large scope in which analysis of situation or data requires a review of identifiable factors. Exercises judgment within defined procedures and practices to determine appropriate action.
Highly-skilled with extensive proficiency.?? Works on complex projects related to field.
Works under minimal supervision. Wide latitude for independent judgment.
Advanced degree preferred. Six to nine years experience post undergraduate degree. One to two year work experience post education
