Position Summary
The Shpargel Laboratory within the UNC Genetics Department is seeking a Research Technician with a background in molecular biology. Histone post-translational modifications can impact expression of large sets of genes during cellular differentiation. The enzymes that catalyze these modifications are mutated in human craniofacial and skeletal disorders. The focus of this study involves utilizing genetic, molecular, and biochemical applications to study the mechanisms by which mutation of histone modifying enzymes regulate bone and cartilage development. Current projects are examining the roles of these enzymes in balancing stem cell biology with osteoblast and chondrocyte differentiation. Minimum Education and Experience Requirements

Demonstrated possession of the competencies necessary to perform the work.

Required Qualifications, Competencies, and Experience
Candidate should possess laboratory technical experiences which includes some combination of the following: mammalian cell culture, PCR, genotyping, chromatin immunoprecipitation, and tissue histology.
Preferred Qualifications, Competencies, and Experience
The project will analyze molecular mechanisms of bone and cartilage development in mice carrying mutations in histone modifying enzymes. Experience in molecular biology, mammalian cell culture, or mouse developmental biology is preferred. This project will collect mutant cell culture or dissected mouse tissues of interest, perform genotyping, and prepare samples for downstream next generation sequencing to analyze gene expression (RNA-seq) or histone profiles (ChIP-seq). The project will utilize a variety of genetic approaches to mutate and assay gene function in cell culture. We will be performing CRISPR screens and proteomics to identify factors that co-function with histone modifying enzymes.
