Research Specialist/Post-Doctoral Researcher in Plasmid DNA Engineering for Lipid Nanoparticle-Mediated Drug Delivery

Overview: We are seeking a highly motivated and skilled individual to join our dynamic team at the Brenner Bioengineering Lab (UPenn) as a Research Specialist or Post-Doctoral Researcher. The successful candidate will play a pivotal role in advancing our research efforts focused on the development of lipid nanoparticle (LNP)-mediated drug delivery systems for various disease indications. Specifically, this role will involve designing, engineering, and optimizing plasmid DNA constructs for efficient delivery via lipid nanoparticles to produce therapeutic proteins.

Responsibilities:

  • Design and engineer plasmid DNA constructs tailored for efficient delivery via lipid nanoparticles.
  • Explore various strategies to enhance the safety and efficacy of DNA-LNP delivery, including but not limited to:
  • Modifying promoters to regulate gene expression levels cell-specifically.
  • Optimizing DNA sequence for optimal gene expression and minimal immunogenicity
  • Incorporating nuclear localization signals (NLS) peptides to enhance nuclear translocation.
  • Developing LTR constructs for improved gene expression.
  • Collaborate with interdisciplinary team members to integrate engineered plasmid DNA constructs into lipid nanoparticle formulations.
  • Conduct in vitro and in vivo studies to evaluate the efficacy and toxicity profiles of engineered DNA-LNP formulations.
  • Analyze and interpret experimental data and contribute to the preparation of research publications and grant proposals.
  • Stay abreast of the latest developments in plasmid DNA engineering, lipid nanoparticle technology, and drug delivery strategies through literature review and scientific conferences.

    Qualifications:

  • Ph.D. in Molecular Biology, Bioengineering, Pharmaceutical Sciences, or a related field with a strong emphasis on plasmid DNA engineering.
  • Extensive hands-on experience in designing and constructing plasmid DNA vectors, including knowledge of molecular cloning techniques and vector design principles.
  • Proficiency in molecular biology techniques such as PCR, DNA sequencing, and plasmid purification.
  • Familiarity with lipid nanoparticle-based drug delivery systems is desirable but not mandatory.
  • Excellent analytical and problem-solving skills, with the ability to independently design and execute experiments.
  • Strong written and oral communication skills, demonstrated by a track record of scientific publications and presentations.
  • Ability to work collaboratively in a team-oriented research environment.
