Guarnerio Lab

Our lab's goal is to understand the interactions between highly aggressive cancers and the non-malignant cells of the microenvironment in order to identify molecular vulnerabilities and develop novel therapies, especially in sarcoma and leukemia. We work in a highly collaborative environment incorporating functional genomics, proteomics and biochemistry approaches that include single-cell and spatial single-cell transcriptomics, mass-cytometry, Hyperion imaging, and state of art mass spectrometry technologies.

We are seeking highly motivated individuals interested in being involved in research projects in one of the following areas:

Sarcoma Microenvironment

Soft-tissue sarcomas comprise a heterogeneous group of aggressive tumors that originate from cells belonging to the mesenchymal lineages. Compared to epithelial carcinomas, sarcomas have been less studied, and lack effective therapeutic alternatives to radiation and chemotherapy. With the aim of identifying new and more effective therapies for these tumors, the lab is employing syngeneic mouse models (Guarnerio et. al., Cancer Discovery 2015 ) to study immune and non-immune cells of the tumor microenvironment through single-cell transcriptomics and functional genomics.

Cancer-associated circRNAs in the microenvironment

We have previously discovered tumor-associated circular RNAs (circRNAs) that act as oncogenes in the process of tumorigenesis (Guarnerio et. al. Cell 2016 ). More recently, we have observed that circRNAs can impact the tumor cells’ “secretome” and ultimately the composition of the tumor microenvironment (Guarnerio et. al., Cell Res 2019 ). Using novel biochemistry techniques refined in our lab, we are studying the unique mechanisms by which tumor-associated circRNAs define the immune and non-immune composition of the tumor mass.

The ideal candidate should have:

  • Technical proficiency in cell culture and hands on mouse work (s.c, i.v, i.p. injections).
  • Some experience in flow and/or mass cytometry.
  • Individuals with training in cancer biology and functional genomics techniques (i.e. shRNA and CRISPR/Cas9 tools) are particularly encouraged to apply.
  • Training in statistics or programming is a big plus but not required.
  • Excellent oral and written communication skills.
  • Ability to perform and prioritize multiple simultaneous tasks.
  • Candidate should be industrious, highly collaborative, and enthused by a vibrant team environment.

Responsibilities:

  • Help to organize and manage experiments in a dynamic environment
  • Execute, and critically interpret experiments and present data in a cogent and compelling manner
  • Present data at lab meetings
  • Under the direction of the PIs, assist in drafting study protocols

Education: BS or MS Degree

Experience:
