The organizational unit is the Marsico Lung Institute/Cystic Fibrosis Center at UNC AT Chapel Hill. This position is located in the Chen lab at MLI/CF Center. The Chen lab use genetic approaches to conduct mechanistic studies relevant to the pathogenesis of chronic pulmonary diseases characterized by airway infection, inflammation, and overproduction of mucus, such as cystic fibrosis (CF), idiopathic pulmonary fibrosis (IPF), and the COVID-19, utilizing in vivo and in vitro culture models relevant to each disease. Specifically, we use both transgenic/knockout mouse models, primary normal and diseased human bronchial epithelial cells coupled with CRISPR-Cas9 technology to address the key roles of transcription factors in respiratory epithelial cells that regulate secretions of mucus and ion/water, mucociliary clearance and innate host defense that are highly relevant to pathogenesis of chronic muco-obstructive lung diseases and SARS-CoV-2 infection.
Position Summary
The primary purpose of Research Technician position is to assist in the following:
1) Routine lab management and purchasing/receiving
2) Performance of experimental and phenotyping protocols in laboratory mice
3) Management the mouse colonies
4) Cell line and primary cell culture
5) mRNA and protein analysis
6) Generation of data reports and analyses
Scientific techniques include but are not limited to:
* DNA isolation and PCR to genotype and maintain mouse colonies;
* Survival and non-survival surgeries to perform harvest of bronchoalveolar lavage, collection of tracheal and lung tissues;
* Airway targeted instillation of reagents and pharmacologic treatments;
* Intraperitoneal injection;
* Monitoring of survival and health status;
* Cell culture and molecular biology techniques including mRNA and protein isolation, Taqman assays, western blot and immunohistochemistry.
Key components of the Research Technician role include daily lab maintenance, data entry, record keeping, sample cataloging and archiving, and compliance with animal handling, health and safety procedures.
Minimum Education and Experience Requirements

Demonstrated possession of the competencies necessary to perform the work.

Required Qualifications, Competencies, and Experience
Our ideal candidate for this role has a good command of general laboratory procedures along with the following attributes:
* Willingness to work with laboratory mice and be able to work with non-survival dissection and surgery to mice;
* Excellent manual dexterity;
* Effective communication skills;
* Ability and willingness to coordinate with lab members on specialized research techniques, and collaborations that positively impact projects and improve efficiency of experimental processes;
* Willingness to follow established lab safety procedures, proficiency in using Excel software and diligence about maintaining laboratory records.
Preferred Qualifications, Competencies, and Experience
Preferred qualifications include:
* Bachelor’s degree or higher in physiology, molecular biology, genetics, biology, biochemistry or related disciplines
* Ability to present research updates and provide feedback on other team members research in weekly interactive lab meetings
* Familiarity with data analysis software routinely used in the laboratory setting (GraphPad Prism) and the ability to query scientific literature web-based resources (PubMed)

Ability to move 25 lbs with or without accommodation; no known allergies to laboratory animals (mice, rats).
