The mission of our Duncan lab is to conduct basic and clinical research in the areas of gonorrhea immunology, pathogenesis, and molecular epidemiology using both classical approaches (ELISA, western blot, flow cytometry) and multi-omic approaches (proteomics, RNASeq, single-cell RNAseq, microbiome analyses).
Position Summary
The primary purpose of the Lab Animal Coordinator position is to conduct independent scientific experiments under the direction of Dr. Duncan, act as the animal coordinator for the laboratory, manage, and/or assist in the various maintenance and administrative functions of the laboratory.
Scientific experiments will include the following areas of study and techniques:
* Deployment of animal models of infectious diseases, particularly the N. gonorrhoeae vaginal infection model, including the evaluation of murine estrous cycle, preparation of bacterial inocula, quantitative bacterial culture, and cellular analysis of vaginal swabs and lavages
* Learn and perform cell culture of human and mouse cells to study immune responses to Neisseria gonorrhoeae with flow cytometry, cell, sorting, RNAseq, single-cell RNAseq, protein isolation, ELISA, Western blotting, immunoprecipitation, and plasmid prep
* Learn and perform sequencing library generation and maintain relevant clinical specimens, lab samples and materials
Lab Animal Coordinator duties include
* Certification as a laboratory animal coordinator by UNC’s IACUC, including serving as primary emergency (24h) contact for lab IACUC protocols
* Ensure IACUC protocols are submitted and updated when necessary, along with updated and current chemical list
* Training others in techniques used in the mouse model of gonorrhea infection
* Ensure all agents used in animals are maintained in appropriate logs and storage requirements.
* Ensures personnel on protocol are current with training and new employees are properly trained
* Ensure animal welfare compliance requirements
Management and administrative duties include
* Ordering of laboratory reagents and maintenance of laboratory materials stocks
* Ensure that purchase records are maintained, and necessary documentation provided to accounting.
* Training of incoming lab personnel in laboratory techniques
* Biobanking and maintaining specimen collections, including freezer organization
* Maintain Laboratory Safety Plan according to requirements of the Department of Environment, Health and Safety for Duncan/Hobbs laboratories
Minimum Education and Experience Requirements
Bachelor’s degree in a discipline related to the area of assignment; or equivalent combination of training and experience. All degrees must be received from appropriately accredited institutions.
Required Qualifications, Competencies, and Experience
Experience with the following assays: ELISA, PCR and qPCR, DNA and RNA extraction and purification. Previous experience maintaining and updating lab protocols. Previous experience maintaining repositories of human clinical specimens. Ordering and inventory of reagents and equipment is also required. Animal handling techniques as well as lab techniques
Preferred Qualifications, Competencies, and Experience
Has performed the following tasks and responsibilities related to in vivo mouse experimentation:
- Performed in-vivo mouse experiments with mouse models of bacterial infection, preferably genital infections such as gonorrhea or chlamydia
- Has tested drug compounds or vaccines in mice and monitored mouse survival rates
Has experience with the following laboratory procedures and activities:
- Flow cytometry
- Cloning and Plasmid preparation
- Streaking, inoculating and incubation bacterial culture in liquid media or on agar plates
- Mammalian cell cultures, cell counting and cell isolation (particularly primary macrophage & lymphocytes)
- Has training in Radiation Safety, conducted swipe tests, and maintained radiation inventories
- Has experience with Laboratory Safety Procedures and/ service as laboratory Safety Officer
- Lab organization, management, and administrative duties,
-Experience with Graphpad Prism and Microsoft Office Suite software (word, powerpoint, and excel)
-Manuscript experience.
