Summary

Note: This appointment is part-time at 75% effort/30 hours per week.  

The laboratory of Daniel Klionsky at the Life Sciences Institute is looking for an Assistant Research Scientist to oversee a research project to study the role of autophagy in organelle homeostasis in fungi. Major goals include the development of an assay to monitor Golgi biogenesis and turnover to identify components involved in selective degradation of secretory pathway organelles under basal and stress conditions. The approach will require expertise in cell and molecular biology along with molecular genetics and considerable experience with various microscopy techniques. The project is anticipated to require recombinant DNA techniques (e.g., cloning, PCR), strain construction (e.g., the generation of knockout and point mutants, fluorescent tagging of specific proteins), and protein analyses (e.g., immunoprecipitation, western blot, microscopy). Due to differences in the spatial arrangement of the secretory pathway you will have experience working with both S. cerevisiae and K. phaffii/P. pastoris. In addition to planning and performing experiments, you will oversee research performed by graduate and undergraduate students and will work collaboratively with other members of the laboratory. You will report directly to Dr. Klionsky and will work on-site.

Responsibilities*

Experimental responsibilities will include:

• Design experiments independently and in collaboration with other lab members
• Generation of yeast strains expressing mutant and fluorescently tagged proteins
• Development of microscopy-based assays for monitoring changes in organelle status
• Carrying out assays for macroautophagy
• Solving experimental problems that arise during any of the responsibilities

Non-experimental responsibilities will include:

• Provide consultation and advice to lab members in areas of expertise, particularly microscopy
• Maintain clear laboratory notebooks
• Provide detailed documentation so experiments can be replicated
• Follow laboratory best practices for safety and quality
• Help manage projects
• Assist or collaborate with the PI in preparing manuscripts and grants

Required Qualifications*

• Ph.D. in molecular biology, cell biology, biochemistry, biophysics, or related discipline
• Minimum 5 years of experience of postdoctoral work related with S. cerevisiae and K. phaffii/P. pastoris
• Minimum 5 years of experience in cell and molecular biology techniques in organelle biogenesis and the secretory pathway
• Minimum 5 years of experience in confocal, fluorescence and electron microscopy using yeast as the model system
• Minimum 3 years of experience mentoring students and research staff
• Strong publication record in relevant work
• Prior experience working in a collaborative environment
