Contact term: This is a 6 month full-time position on Crick terms and conditions of employment.

Summary
This project is to develop assays which can be used in high-throughput screening (HTS) for drug discovery programmes to target autophagy. The successful applicant will develop proximity-based assays to measure protein-protein interactions suitable for compound screening in mammalian cell lines. These protein-protein interactions are part of the mechanisms underlying the formation of autophagosomes. The project will employ state-of-the-art imaging techniques, HTS screening approaches and will require monitoring protein localization and biochemical assays. The applicant will be working as part of a team dedicated to understanding the early stages of autophagy and with a team supported by Cancer Research UK.

This position would suit a current Crick staff member coming to the end of their contract, or would also be available as a secondment.

The Research Group
Dr Tooze's laboratory focuses on the molecular basis underlying autophagy (self-eating). Details of research projects currently being undertaken can be seen at:

The research aims to create assays suitable for high-throughput screening, however the biology underlying the protein-protein interactions can also be explored with the team dedicated to understanding the autophagic pathway. This work will also contribute to the research funded by an ERC grant.

Key experience and competencies
The post holder should embody and demonstrate our core Crick values: bold, imaginative, open, dynamic and collegial. The successful applicant should have the ability to deliver results in a timely manner, and a thorough understanding of experimental design, appropriate controls and reproducibility is essential. The successful applicant should also be able to handle large numbers of samples, and work with instrumentation designed to facilitate high-throughput screening, and manage data using appropriate software. Finally, attention to detail is essential for the development of robust assays.

Essential

Good knowledge and experience in mammalian cell biology
Technical expertise in mammalian cell based approaches, light microscopy, and biochemistry
Experience in expression of proteins in mammalian cells
Track record of excellent record keeping
Experience of experimental design
Ability to work independently and also capable of interacting within a group
Desirable

Experience in cell based high-throughput assays
Experience in fluorescence-based assays, FRET, FLIM
Experience with confocal imaging
