Offer Description

KEY WORDS: genome editing, CRISPR/Cas9, safety, gene therapy, DNA repair

Principal Investigator: Marta Olejniczak, PhD, Associate Professor of IBCH PAS

Research topic: “Assessing efficacy and safety of genome EDITing approaches for Sickle Cell Disease

Research will be carried out within the EDITSCD project (https://editscd.eu/ ) funded by European Commission (2022-2027). The EDITSCD consortium is coordinated by Annarita Miccio at the Institut Imagine (France). The other international partners of the consortium are the Ospedale San Raffaele SRL (Italy), the Universitaetsklinikum Freiburg (Germany), the Bar Ilan University (Israel), the Inserm (France), the ETH Zurich (Switzerland), AstraZeneca AB (Sweden) and EATRIS ERIC (Netherlands). ICHB will join this ongoing project with its own budget under the Hop-on Facility (EDITSCD-HOP-ON).

Project description: Sickle cell disease (SCD) is a group of inherited red blood cell disorders that affect hemoglobin. A single amino acid substitution in the beta-globin chain of adult hemoglobin causes red blood cell sickling, hemolysis, vaso-occlusive crises, and multi-organ damage. Transplantation of autologous, genetically modified hematopoietic stem/progenitor cells (HSPC) represents a therapeutic option for SCD. The EU-funded EDITSCD project aims to understand the molecular and cellular mechanisms underlying SCD HSPC dysfunctions and evaluate the impact of genome editing approaches on SCD HSPC. One of the EDITSCD project objectives is to establish efficient and safe genome editing approaches to correct the SCD mutation. As part of this project, ICHB will develop and validate a new HDR-based method for precise correction of the beta-globin gene.
