OFFER DESCRIPTION

Germline mutations in the transcription factor GATA2 result in bone marrow failure with a high predispostion for leukemia. We have modeled this disease in zebrafish by mutating Gata2b, affecting both embryonic and adult hematopoiesis. To be able to establish future gene therapy treatment options for these patients, we need to understand if adult restoration of the Gata2 deficiency is sufficient to abrogate disease ontogeny. For this we have generated a mutant that, upon CRE activation, reexpresses Gata2b from its endogenous locus. The goal of this project is to investigate if reexpression of Gata2b restores the adult leukemia phenotype. The project involves embryonic and adult zebrafish manipulation, flowcytometry analysis and state-of-the-art molecular analyses. ]
