Offer Description
Signal Transducer and Activator of Transcription 3 (STAT3) is a transcription factor activated by a wide range of cytokines, growth factors and hormones. Mutations of STAT3 underlie a wide spectrum of disorders leading to immunodeficiency, autoimmunity and malignancy. Heterozygous germline loss-of-function (LOF) mutations cause autosomal dominant-Hyper-IgE Syndrome (AD-HIES). Despite the extensive characterization of STAT3 functions at molecular and cellular level, several aspects, relevant to the clinic, remain to be elucidated.The research program has the following aims:identification of patients with STAT3-related disease and clinical and immunological assessment;deconvolution of innate and adaptive immune cell complexity in patients with STAT3-related inborn errors by immunophenotyping and scRNAseq;dissecting new roles for STAT3 and STAT3-target genes underlying regulation of effector functions in immune cells.
