The Human Technopole Foundation is searching for a Senior Research Technician to join the Automated Stem Cell and Organoid Facility (ASCOF). The aim of the Facility is to enable patient-specific disease modelling at scale by translating, benchmarking and standardising laboratory protocols into automated workflows, integrating state-of-the-art robotic and bioengineering platforms through a highly multidisciplinary approach. Operating in close connection with the Genomics, Advanced Imaging, FACS and Electrophysiology facilities and support units, ASCOF develops transformative technologies to realize the Human Technopole vision of high definition biomedicine through the use of increasingly sophisticated organoid systems at single cell multiomic resolution, with a strong focus on brain diseases and densely phenotyped clinical cohorts.

The successful candidate will report to the Head of Automation and will have the opportunity to work in a multidisciplinary environment to develop automated protocols in the following areas: reprogramming of somatic cells into induced pluripotent stem cells (iPSCs), brain organoids and gene editing.

S(he) will have the following main responsibilities:

Develop and optimize cell culture protocols in the area of stem cell neurobiology, combining cutting-edge methods from molecular biology, cell biology and organoid technology, biochemistry and histology
In synergy with the automation team, develop innovative solutions to adapt conventional cell culture routines to automated platforms
Generating/importing new tools and protocols
Supporting the head of automation in setting up the Facility resources, participating in key aspects of general facility management and organization (e.g., managing inventories and stocks of samples, reagents, equipment and materials)
Job requirements
At least 5 years of relevant work experience;
Track record in the following areas:
Advanced cell culture experience with human pluripotent stem cell lines, organoids and primary neural cell cultures
Familiarity with the complete portfolio of CRISPR based genome editing techniques: from Knockout line generation to engineering of specific mutations and/or reporters, as well as gene expression manipulation through CRISPRi and CRISPRa, and CRISPR screening approaches
Molecular Biology (DNA and RNA manipulation, cloning, sequencing, RT-PCR and library preparation for next-generation sequencing applications, single cell manipulation, FACS)
Familiarity with Protein Biochemistry methods (immunoblotting and immunoprecipitation techniques, preferably also recombinant protein expression, purification and in vitro reactions), Tissue processing and Histology (preferably on mammalian brain)
Experience and skills in performing repetitive and specialized laboratory experimental procedures. Experience following experimental protocols;
Fluency in English – HT is an international research institute;
