Exciting opportunity in Polycomb Research in Children’s Cancer

  • Reports to: Prof O’Sullivan.
  • Salary: Apppointment will be made according to the Irish Universities Association (IUA) post-doctoral level 1(PD1) in line with government pay policy [41,209-47,477 per annum, in accordance with the candidate’s level of experience].

  • Start date: The successful candidate will be expected to take up the position as soon as possible.
  • The O’Sullivan research Laboratory Group has an international reputation in research on paediatric renal tumors and specifically those showing perturbations of chromatin regulation. The particular project is on the polycomb repressor complex 1.1 (PRC1.1), notably the BCOR gene, and its role in a rare and aggressive paediatric cancer known as Clear Cell Sarcoma of Kidney (CCSK). 

    We have completed a large body of work on the genetics of this tumour and the current project builds on that with an investigation into mechanistic effects of internal tandem duplication (ITD) of BCOR on PRC1.1 complex assembly and function. A novel finding has emerged through the application of IP-Mass spec, Gel filtration with Western blotting of our cell line models for this tumor, and molecular dynamic modelling. Current ongoing work includes RNA sequencing, ChIP sequencing and pharmacological manipulations of the cell lines to complete an in-depth and multi-faceted investigation into the effects resulting from the BCOR-ITD. The project is already quite mature and high impact publication is planned within months. 

    Your duties will include:

    • Taking on responsibility for the timely completion of some of these ongoing experiments to the highest standards including quality and safety considerations.
    • Troubleshooting experimental difficulties and /or seeking assistance where needed.
    • Maintaining clear laboratory notebook records.
    • Regular clear communication and liaison with the PI in relation to progress, organisational plans (including ordering and maintaning necessary stocks for timely completion of experiments) and in relation to results being accrued and mutual interpretation thereof.
    • Contribution (intellectual as well as technical) to a multi-member team analysing the sequencing results (RNA- and ChIPSeq) – we have excellent bioinformatics support in place already also.
    • Contribution to preparation of the manuscripts including production of figures for publication.

    Essential qualifications include MSc or PhD in cell biology, molecular biology or genetics.

    Necessary Laboratory Skills include:

    Cell culture, Nucleic Acid and Protein extraction, PCR, Western blotting.

    Desirable laboratory skills include:

    ChIP, ChIPSeq and some bioinformatics knowledge.

    Personal skills required:

    Excellent communication skills, proficiency in English, both spoken and written, Ethical and responsible approach to work with complete transparency at all times.

    Enthusiasm, dedication and high-level of motivation to complete experiments to the highest standards.

    Ability to work independently but also maintaining a great team approach.
