The Research Unit Translational Immunology (TUM) and the Institute of Lung Health and Immunity (LHI; Helmholtz Munich) joined forces to better understand and cure lung diseases associated with a dysregulated STAT3 signaling using CRISPR-mediated gene editing. The proposed project focuses on STAT3 gene correction in lung cancer and STAT3-hyper-IgE syndrome (STAT3-HIES), which is a monogenic inborn error of immunity with chronic destructive lung disease. To correct the underlying disease-causing STAT3 mutations we established CRISPR/Cas9-mediated base editing (Eberherr et al. CRISPRJ). Your responsibilities:
 To develop an innovative technique to correct lung diseases as part of an interdisciplinary team
 To perform gene editing of human lung cancer cells and iPSC-derived alveolar epithelial cells
 To assess efficiency and safety of the delivery method and the performed CRISPR-mediated gene editing

Your profile:
 PhD in one of the life sciences (preferably pharmaceutical studies), MD or equivalent work experience in the wet lab
 A strong background in cellular delivery of nucleic acids or proteins including formulation development and cell culture technique is favorable
 Ideally you have experience in gene editing (including gRNA design and validation)
 Experience in downstream analysis including immunofluorescence staining, flow cytometry, western blot analysis, quantitative real-time PCR, Sanger sequencing, high-throughput sequencing, in silico analysis of editing efficiency, cell viability analysis is a plus
 The highly motivated candidate must be able to work independently however we greatly seek team player mentality

 The project is initially limited to two years. An extension of the project is planned.
