Offer Description

The post-doctoral researcher will work in the Laboratoire Analyse, Modélisation et Matériaux pour la Biologie et l'Environnement (LAMBE, CNRS UMR 8587, Université Paris-Saclay, site Evry Univ.), analyzing bioactive polysaccharides glycosaminoglycans (GAGs) of synthetic and biosynthetic origin and characterizing their multivalent assemblies on macromolecular platforms. These structural and functional characterizations will be based on the use of several analytical chemistry and biochemistry methods, in particular mass spectrometry (MS) and couplings with separative methods. This work forms part of a national research program (ANR) over the period 2022-2025, aimed at using these multivalent platforms to inhibit excessive collagenase activity, which is implicated in osteoporosis-type bone degeneration. The project will be carried out under the scientific responsibility of the theme leader. The various aspects of the research project include - implementation of MS MALDI TOF/TOF and electrospray (ESI) analysis methods with different fragmentation modes for the detection and structural characterization of GAGs of biosynthetic or chemically synthesized origin. - coupling of separative methods with MS - implementation of enzymatic GAG transformation processes - structural characterization of oligosaccharides by these methods of oligosaccharides and glyco-conjugates; comparison with other analytical methods (chromatography, electrophoresis, NMR, mass spectrometry, among others), - Establishment of databases of structural signatures of oligosaccharides of varying size, sequence and organization. - implementation of affinity methods for the analysis of bio-interaction properties with target enzymes.

- analysis of their attachment to different macromolecular platforms
- Complementary structural analysis of oligosaccharide libraries (chromatography, electrophoresis, NMR, etc.)
- Detection and characterization of biomolecular interactions using affinity techniques (affinity electrophoresis, SPR)
- Processing analytical results for publication,
- Participate in the writing of scientific articles.
- Apply health and safety rules in the field.

LAMBE is a CNRS joint research unit (UMR 8587) at the Université Paris-Saclay, Evry Univ. site, whose activities come under the "Chemistry and Life" and "Physical, Theoretical and Analytical Chemistry" sections of the CNRS Institute of Chemistry (INC). Its research themes mainly concern the structure/reactivity of biomolecules, particularly in the field of glycosciences. To this end, the laboratory exploits and develops methods based, among others, on mass spectrometry and its couplings (LC, CE, SPR). The Laboratory has developed solid expertise and know-how in the analysis and structural elucidation of biomolecules, in particular complex carbohydrates such as glycosaminoglycans, by MS and coupled methods. The laboratory operates a platform of seven mass spectrometry instruments. All the equipment and expertise required for the project are available in the laboratory.

Eligibility criteria

The candidate must have a PhD in a field related to analytical/bioanalytical chemistry (separation methods, chromatography, mass spectrometry and coupled methods applied to biomolecules) and/or biochemistry (glycosciences, protein chemistry). Solid knowledge is required of analytical chemistry (preparation, purification & detection) and structural characterization methods, in particular mass spectrometry. Knowledge of biochemical and biophysical bio-structural methods is desirable. The candidate will be motivated to take on scientific challenges. The candidate will demonstrate good writing skills, with a good command of written English, and the ability to publish and promote research work. The candidate will be able to work as part of a team and network with partners.
