Offer Description
Project summary: Aging of the hematopoietic system results from the deterioration of the pool of hematopoietic stem cells (HSCs) that drift in response to environmental stress over time. Inflammation as well as extrinsic signals from the hematopoietic niche are likely to contribute to this deterioration by modulating the transcriptomic and epigenetic profiles of HSCs. Notably, there is a sexual dimorphism in HSC aging, and our working hypothesis is that by understanding the molecular basis of these differences, we may be able to uncover regulatory networks important in aging. In this project, we will explore the differences in gene expression and epigenetics between male and female mouse HSCs. We will identify the molecular mechanisms underlying these differences and test their potential in preserving HSC function, in order to propose solutions for maintaining a high-performing hematopoietic system over time.
Mission: The candidate will be responsible for developing single-cell multi-omics analysis approaches (sc-RNAseq, scATACseq, 10X Genomics) to identify and characterize markers of aging differences between male and female HSCs, and to functionally test the effect of these markers on HSC aging.
