Afin d’évoluer dans notre environnement volatile, nous faisons constamment appelle à notre sens de l’odorat. Chez les mammifères, il est désormais bien établi que les récepteurs olfactifs jouent un rôle important dans l'olfaction et d'autres processus développementaux et physiologiques importants, dont certains sont associés à des problèmes de santé graves. Le mécanisme moléculaire exact par lequel les récepteurs olfactifs accomplissent leur fonction est méconnu et nécessite des informations à l’échelle atomique qui ont fait largement défaut. Il s’agit d’une famille de protéines peu étudiée, bien qu’elle représente près de la moitié de la grande famille des récepteurs couplés aux protéines G, principalement parce qu’il n’existait, jusqu’à récemment, aucune structure expérimentale de récepteur olfactif de mammifère. Le manque d’expression des récepteurs olfactifs dans des modèles de cellules eucaryotes utilisés pour l’expression des protéines membranaires recombinantes constitue le principal obstacle pour relever ce défi. Nous avons réussi à débloquer leur expression en modèle cellulaire grâce à la construction de récepteurs « consensuels » qui résument l’ensemble des propriétés des récepteurs olfactifs humain, nous permettant cette année de publier la première structure d’un récepteur olfactif. Le projet s’orientera vers l’étude en cytométrie en flux de l’expression de constructions innovantes de récepteurs olfactifs à la surface de cellule HEK ainsi que la mesure de leur réponse à des molécules odorantes d’intérêt pour la parfumerie et le drug design. La visualisation et caractérisation des structures de ces récepteurs et de leur liaison avec les molécules odorantes se fera en modélisation moléculaire. Cette thèse fera donc intervenir des méthodes de biochimie, culture cellulaire et modélisation moléculaire. Co-direction entre Claire de March (principale) et Ewen Lescop.

In order to navigate our volatile environment, we constantly rely on our sense of smell. In mammals, it is now well established that olfactory receptors play an important role in olfaction and other crucial developmental and physiological processes, some of which are associated with serious health issues. The exact molecular mechanism by which olfactory receptors accomplish their function is not fully understood and requires atomic-scale information, which has been largely lacking. This is a relatively understudied family of proteins, even though it represents nearly half of the large family of G protein-coupled receptors, primarily because, until recently, there were no experimental structures of mammalian olfactory receptors. The lack of expression of olfactory receptors in eukaryotic cell models used for expressing recombinant membrane proteins is the main obstacle in addressing this challenge. We have succeeded in unlocking their expression in a cell model through the construction of "consensus" receptors that summarize all the properties of human olfactory receptors, allowing us to publish the first structure of an olfactory receptor this year. The project will focus on studying the expression of innovative olfactory receptor constructs on the surface of HEK cells using flow cytometry, as well as measuring their response to odorant molecules of interest for perfumery and drug design. The visualization and characterization of the structures of these receptors and their binding with odorant molecules will be conducted through molecular modeling. This thesis will therefore involve methods of biochemistry, cell culture, and molecular modeling.

Minimum publication during the PhD: one review article, one protocol, and two research articles.

Le projet étant transdisciplinaire, la candidate ou le candidat devra avoir des compétences dans au moins l'un des domaines cités dans le projet, à savoir la biochimie, la biologie cellulaire, la modélisation moléculaire ou la science des odeurs. Elle ou il devra montrer de l’enthousiaste à l'idée d’acquérir des compétences dans les domaines non maitrisés pour l'instant. La maitrise de l'anglais est un plus.
As this project is interdisciplinary, the candidate should have expertise in at least one of the fields addressed in the project namely biochemistry, cell biology, molecular modeling, or the science of odors. They should also be enthusiastic about acquiring knowledge in areas that are not currently mastered. Mastering english is a plus.
