Our Research and Environment
The overarching goal of the research in the Treebak Group is to define molecular and physiological mechanisms by which dietary compounds, weight loss, and physical activity improve insulin sensitivity to prevent and treat obesity and type 2 diabetes. We are particularly interested in defining mechanisms linking cellular NAD+ to metabolic health. In particular, we aim to understand the role of the NAD+ generating and consuming pathways for maintaining tissue function and adaptability in response to metabolic stress. We employ innovative preclinical models to manipulate NAD-related metabolites cell type-specifically and determine functional implications. We have developed a number of novel and unique experimental models and techniques related to in vivo animal research. Moreover, we apply unbiased omics approaches and analytical pipelines in collaboration with key national and international partners to identify responsible mediators and to determine genotype/phenotype interactions. We are highly motivated and self-driven with a passion for metabolic research. Job Description
Insulin resistance in peripheral tissues such as skeletal muscle and adipose tissue is characterized by an inability of the target tissue to appropriately modulate glucose disposal in response to a specific level of insulin. Insulin resistance precedes the development of type 2 diabetes (T2D) and is a consequence of ectopic lipid deposition in insulin responsive tissues. NAD+ has emerged as an important cofactor linking energy status with adaptive cellular and organismal responses, and it serves as a substrate for multiple enzymes involved in the regulation of energy metabolism. Low cellular NAD+ levels could therefore prompt the development of many of age-related disorders, including insulin resistance. Skeletal muscle act as an important sink for glucose clearance under insulin-stimulated conditions, and insulin signaling to glucose disposal in skeletal muscle in the context of insulin resistance has been studied intensively over many years. We have preliminary data from quantitative mass spectrometric analysis of skeletal muscle that show key proteins in the insulin signaling cascade are ADP-ribosylated. Since ADP ribosylation (ADPr) is mediated by NAD+-consuming ADP-ribosyltransferases, this indicates a yet unrecognized interplay between insulin signaling and cellular NAD+ metabolism. Our hypothesis is that ADPr plays an essential role to regulate insulin sensitivity in skeletal muscle, and our over-arching aim is to determine the role of this posttranslational modification (PTM) for maintaining cellular glucose homeostasis and insulin action. Your tasks would be to define and compare insulin-induced ADP ribosylomes of insulin sensitive and resistant skeletal muscle, and to identify key modifications that are important for insulin signaling to mediate glucose disposal. We strive to understand the molecular basis for ADPr in skeletal muscle insulin resistance, and we are looking for candidates who are motivated to integrate data available from high-resolution mass spectrometry-based proteomics analyses with approaches in in vivo physiology and molecular biology.

The fellowship is aimed at early career researchers with a basic science background. We are particularly interested in candidates who are familiar with integrative research approaches within the broad area of basic metabolic research with an application towards human pathophysiology.

Required qualifications:

  • PhD in Biology, Biochemistry, Molecular Biology, Molecular Biomedicine, or related sciences
  • Excellent scientific track record in relation to career stage
  • Experience in the study of animal physiology, specifically related to in vivo and in vitro metabolic studies using transgenic and knockout models
  • Experience with cell cultures
  • Prior experience in research fields related to insulin signalling/sensitivity, NAD metabolism or ADP-ribosylation would be a clear advantage
  • An analytical aptitude of devising innovative scientific or technical solutions
  • Prior experience in high-resolution mass spectrometry is not essential, but will be considered an advantage
  • The candidate should be able to conduct independent and creative research, demonstrate scientific productivity, and show good inter-personal and communication skills
  • Strong motivation and excellent scientific skills are essential
  • Excellent English communication skills, both written and oral

The CBMR International Postdoctoral Program is open for applicants that hold a PhD degree awarded from a university outside of Denmark. The PhD degree has to be obtained before October 1, 2021. The program is also open for applicants with a Danish PhD degree who can document at least 12 months full-time research experience from outside Denmark. You are not eligible to apply for the program if you have been employed in a postdoctoral position for more than one year at the University of Copenhagen prior to the commencement of the fellowship.

Terms of Employment
The employment as postdoc is a full time position for 3 years. Starting date is October 1, 2021.
Salary, pension and terms of employment will be in accordance with the agreement between the Danish Ministry of Finance and AC (Danish Confederation of Professional Associations). Depending on qualifications, a supplement may be negotiated.
Non-Danish and Danish applicants may be eligible for tax reductions, if they hold a PhD degree and have not lived in Denmark the last 10 years.
