The Technician is primarily responsible for analyses of SARS-CoV-2 infected samples using quantitative cell imaging, genome-wide analyses, and protein-protein interactions. The successful applicant will be expected to undergo training to work in the Biosafety Level 3 laboratory (BSL3).

Duties

  • Conduct infection and analyses of primary and continuous cell cultures in BSL3 facility;
  • Maintain virus, cell culture stocks and cell culture equipment in BSL2 and BSL3 facilities;
  • Perform molecular biological procedures for the construction and characterization of expression vectors;
  • Participate in large-scale genetic and interactome screens using CRISPR and immunoaffinity purification/mass spectrometry;
  • Analyses of large genetic and protein datasets;
  • Perform experiments involving routine and slightly complex procedures in order to meet research objectives;
  • Modify existing protocols for existing experiments in order to meet research objectives;
  • Consult with principal investigator to develop new experiments and techniques for the research project;
  • Use a variety of lab equipment to conduct experiments and monitor results;
  • Collect and organize data on experiments and conduct preliminary analysis of large scale genetic and interactome screens;
  • Prepare figures for manuscripts;
  • Prepare and maintain a variety of specialized solutions for use in experiments;
  • Perform other duties and responsibilities as requested by the principal investigator.

Qualifications and Experience

  • Minimum BSc, MSc preferred.
  • Minimum of two years lab experience.
  • Experience with mammalian cell culture, molecular biology techniques required for protein expression in bacteria and animal cells.
  • Experience with isolation of primary cell cultures from clinical samples.
  • Experience in protein characterization methods, including polyacrylamide gel electrophoresis, immunoaffinity purification, and immunoblotting.
  • Experience in advanced molecular biology techniques including cloning, quantitative polymerase chain reaction, next-generation sequencing.
  • Experience in immunological techniques including Enzyme-Linked Immunosorbent Assays (ELISA) and flow cytometry.
  • Experience in global proteome analyses and protein-protein interactions.
  • Ability to analyze large data sets and pathway analyses using David, Gorilla, Partek Flow, Cytoscape, and/or Metascape.
  • Experience in super-resolution and confocal immuno-fluorescence microscopy.
  • Experience with image analyses software such as Volocity, Imaris and/or ImageJ.
  • Experience with statistical analyses software such as GraphPad Prism and Excel.
  • Experience in manuscript preparation and oral presentations using Microsoft Powerpoint, Adobe Photoshop or Illustrator, Biorender and/or other software for figure preparation.
