Position Summary:
The Brown lab is focused on developing and applying advanced bioimaging techniques including live imaging methods to diverse biological problems. Projects will include: 1) understanding the fundamental molecular mechanisms that regulate cell migration in normal and diseased states though the regulation of cell-matrix adhesions, 2) developing methods for label free imaging of tissue samples as a diagnostic tool for pathology and 3) the role of cell migration in cancer metastasis in cells and tissue. Methods will include Fluorescence Lifetime Imaging Microscopy (FLIM), Stimulated Emission Depletion (STED) super resolution microscopy, spectral imaging and expansion microscopy.
With a strong focus on interdisciplinary work, the position will involve working on independent research projects and working with researchers using the Advanced BioImaging Facility (ABIF) from diverse fields such as cell biology, cancer biology, biophysics, neuroscience and cellular biomechanics to meet their bioimaging needs. McGill is ranked among the finest research-intensive universities in the world. The ABIF of McGill University has technical capabilities for standard widefield and confocal microscopy as well as advanced imaging such as live cell imaging, total internal reflection fluorescence (TIRF) microscopy, FLIM, STED, lattice lightsheet microscopy, fluorescence correlation spectroscopy (FCS), image processing and analysis, spectral imaging and high content screening. The primary duties will include:
1. Fluorescence Lifetime Imaging Microscopy (FLIM): Work with a collaborative team to develop FLIM image acquisition and analysis protocols and test them for diagnostic purposed on normal and diseased tissues. Apply these FLIM methods to other projects with users in the imaging facility. Develop and apply FLIM methods for live imaging applications.
2. STED Microscopy: Develop STED protocols for sample preparation, image acquisition and image analysis for studying cell-matrix adhesions in fixed and live samples. Focus on maximal resolution in fixed samples including development of expansion microscopy ExM protocols that can be combined with STED imaging protocols and live imaging of adhesion dynamics and spatial and temporal regulation of protein binding and interactions in adhesions.
3. Spectral Imaging: Develop spectral imaging sample preparation, image acquisition and image analysis protocols for diverse applications including multilabelled samples, label-free tissues and physiology probes (e.g. pH, membrane potential, membrane lipid properties).
4. Image Analysis: Develop image analysis workflows in collaboration with the ABIF imaging scientists for the various projects.
5. Data Management: Develop and implement a rigorous image data management and sharing plan including metadata collection in collaboration with the ABIF team. This will be aligned with the Canada BioImaging OMERO Image Data Resource.
6. Keep up on the literature relevant to the project.
7. Apply for fellowship funding opportunities.
8. Write and publish research papers, present work at conferences and publish detailed protocols.
Qualifications:
EDUCATION/EXPERIENCE
Required
PhD in preferred areas including Biophysics, Cell Biology, Cancer Biology, Bioengineering. Microscopy Experience with widefield and confocal laser
scanning microscopy.
Some microscopy experience with FLIM, STED and spectral imaging.
Image analysis experience.
Cell culture, molecular biology, live cell imaging.
Ability to manage a diverse set of obligations on a daily basis with rapidly changing demands and priorities.
Strong interpersonal skills.
Ability to work independently and efficiently but also work well as a part of a team.
Strong verbal and written communication in English.
Would be an Asset
Immunofluorescence labelling techniques.
Familiarity with imaging software: FIJI/ImageJ, Imaris, LASAF, ZEN, OMERO. Experience working in a dynamic environment.
Strong verbal and written communication in French would be an asset.
