Offer Description

One position is available at the University of São Paulo's Medical School (FM-USP) in Brazil to conduct a Post-Doctoral research project which consists of the use of zebrafish (Danio rerio) as an animal model to elucidate the functional effect of allelic variants identified in patients with differences in sexual development (DSD).

Aim

The development of zebrafish models for the characterization and analysis of potentially harmful allelic variants identified by the Sanger method or by large-scale sequencing (genome or exome study) in patients with DSD.

Specific aims

• Large-scale DNA sequencing of patients with XX and XY sex development differentiation phenotype without molecular diagnosis;
• Creation and maintenance of Danio rerio in the Multiuser zebrafish animal facility at FM-USP;
• Introduction of possibly pathogenic variants, linked to differences in sex development, by the CRISPR/Cas9 technique in zebrafish;
• Collection of gonadal tissue samples at different stages of development of zebrafish;
• Extraction of RNA from gonadal tissues of zebrafish;
• Quantitative expression of gene transcripts;
• In situ hybridization and Immunohistochemistry.

Scientific Activities

• Patient selection;
• Large-scale DNA sequencing of patients with XX and XY sex development differentiation phenotype without molecular diagnosis;
• Optimization of protocols and introduction of possibly pathogenic variants.

This opportunity is open to candidates of any nationality. The selected candidate will receive a Post-Doctoral fellowship from the São Paulo Research Foundation (FAPESP) in the amount of R$ 8,479.20 monthly and a research contingency fund, equivalent to 10% of the annual value of the fellowship which should be spent on items directly related to the research activity.
