Determination of the effect of bacteria from compost on plant growth stimulation and pathogen inhibition

Abstract
In the current period, due to expected climate changes, but also public pressure for organic production without the use of synthetic fertilizers and pesticides, more and more attention is paid to microorganisms (mainly bacteria) that are able to support plant growth or act antagonistically against phytopathogens. Bacteria promoting plant growth (Plant Growth Promoting Bacteria - PGPB) form a specific group that can stimulate the growth of plants by various mechanisms, provide them with a higher intake of nutrients, and increase their resistance to abiotic stresses, diseases and pests. In addition to the rhizosphere, above-ground parts of plants (phyllosphere species) and inside plant tissue (endophytic species), PGPB can also be isolated from composts. In the case of PGPB isolated directly from composts, their good survivability is assumed after the application of composts fortified with such bacteria to the soil and the subsequent improvement of the microbiological parameters of the soil quality as well as a positive effect on the composition of the soil/plant microbiome.
Scientific objectives: (1) isolation of bacteria from composts with an emphasis on monitoring morphological and biochemical characteristics, enzymatic activity, utilization of substrates and properties important in terms of plant growth stimulation and the ability to resist abiotic and biotic stress conditions, mainly pathogens (2) analysis of the action of selected PGPB strains in stimulating plant growth and inhibiting the growth of pathogens and inducing systemically acquired plant resistance as a response to the action of model stress biotic (pathogen) environmental conditions. The induced stress will be evaluated based on the production of jasmonic acid, salicylic acid and ethylene and signaling pathways that have a key role in the regulation of plant defense reactions (3) evaluation of the impact of the application of isolated PGPB on the growth of selected plants and on the quality of the fortified compost with PGPB.
Determinácia pôsobenia baktérií z kompostu na stimuláciu rastu rastlín a inhibíciu patogénov
Abstrakt
V súčasnom období, z dôvodu očakávaných klimatických zmien, ale aj tlaku verejnosti na ekologickú produkciu bez používania syntetických hnojív a pesticídov je stále väčšia pozornosť venovaná mikroorganizmom (hlavne baktériám), ktoré sú schopné podporovať rast rastlín alebo antagonisticky pôsobiť proti fytopatogénom. Baktérie podporujúce rast rastlín (Plant Growth Promoting Bacteria - PGPB) tvoria špecifickú skupinu, ktoré môžu rôznymi mechanizmami stimulovať rast rastlín, zabezpečovať im vyšší príjem živín, zvyšovať ich odolnosť voči abiotickým stresom, chorobám a škodcom. PGPB okrem rizosféry, nadzemných častí rastlín (fylosférové druhy) a vo vnútra rastlinných pletív (endofytické druhy) možno izolovať aj z kompostov. Pri PGPB izolovaných priamo z kompostov sa predpokladá ich dobrá prežívateľnosť po aplikácii kompostov fortifikovaných takýmito baktériami do pôdy a následné zlepšenie mikrobiologických parametrov kvality pôdy ako aj pozitívny vplyv na zloženie pôdneho/rastlinného mikrobiomu.
Vedecké ciele: (1) izolácia baktérií z kompostov s dôrazom na sledovanie morfologickej a biochemickej charakteristiky, enzymatickej aktivity, utilizácie substrátov a vlastností dôležitých z hľadiska stimulácie rastu rastlín a schopnosti odolávať abiotickým a biotickým stresovým podmienkam hlavne patogénom (2) analýza pôsobenia vybraných kmeňov PGPB pri stimulácií rastu rastlín a inhibícii rastu patogénov a indukovaní systémovo získanej rezistencie rastlín ako odpoveď na pôsobenie modelových stresových biotických (patogén) podmienok prostredia. Vyvolaný stres bude hodnotený na základe produkcie kyseliny jasmónovej, kyseliny salicylovej a etylénu a signálnych dráh, ktoré majú kľúčovú úlohu v regulácii obranných reakcii rastlín (3) hodnotenie vplyvu aplikácie izolovaných PGPB na rast vybraných rastlín a na kvalitu fortifikovaného kompostu s PGPB.
