Human Technopole (HT) is an interdisciplinary research institute in Milan (Italy) created and supported by the Italian government to develop innovative strategies to promote human health. HT adopts a multidisciplinary and integrated approach combining genomics, computational and structural biology, neuroscience, and data and decision sciences.

The Human Technopole Foundation is looking for one highly motivated postdoctoral candidate with a strong background in biochemistry or organic chemistry of biomolecules to work in the group of Prof. Marino Zerial.

The Zerial Group works on endocytosis - from its molecular mechanisms to implications in cell and tissue organisation. Its interests range from the assembly and functional characterisation of the endosomal fusion machinery to establishing hepatocyte cell polarity and liver tissue structure and function. The Group uses a unique interdisciplinary approach combining biochemical and biophysical methods with advanced light microscopy, cell biology and computer-aided three-dimensional tissue reconstruction to develop novel targeted oligonucleotide therapeutics.

Specific targeting and endosomal escape remain crucial hurdles for the successful implementation of macromolecular therapeutics. The know-how in endocytosis positions the Zerial Group at the leading edge of understanding what drives these limitations and identifying novel ways to overcome these obstacles.

This project aims to translate novel targeting ideas identified into applications for oligonucleotide delivery. It concerns antibody production against identified targets and their attachment to oligonucleotides and nanoparticles. Their uptake and trafficking will be studied in cell lines and primary cells in vitro and mouse models, using quantitative light microscopy and high-throughput screening approaches. The project will be carried out in collaboration with the pharmaceutical industry.

Key tasks and responsibilities

  • Conduct in vivo imaging studies using quantitative light microscopy to monitor the trafficking and distribution of antibody-oligonucleotide conjugates in cells and mouse models.
  • Compare the uptake efficiency between different cell lines and primary cells, providing insights into the potential variations in cellular uptake.
  • Generate reliable data for publication and record all data and results in specified forms (paper and electronic).
  • Contribute to the writing of project-related scientific papers.
  • Assist in the training of PhD students and contribute to the smooth running of the lab.

Job requirements

The candidate should be able to interact with members of an interdisciplinary team of cell biologists, biochemists, computer scientists and clinicians, work in a competitive international environment and possess excellent communication and data presentation skills.

Mandatory requirements:

On the closing date for online applications, the candidate must fulfil all the following conditions:

  • A PhD in life sciences.
  • Documented prior experience in antibody or oligonucleotide modification or organic synthesis of biomolecules, ideally in antibody conjugation approaches.
  • Proficiency in biochemistry and molecular biology methods and protein purification, quantification and detection techniques.
  • At least one first-author publication in a scholarly journal in a field relevant to this application.
  • Fluency in English.

Preferred requirements:

  • A background in membrane biology, endocytosis and cell trafficking would be advantageous.
  • Previous experience in quantitative light microscopy.
