Offer Description
A two-year postdoctoral position is available in the laboratory of the Pr. Benveniste and Pr. Allenbach in the Center of Research in Myology (U974 Sorbonne Université-INSERM).
The project is fully funded by the ARC foundation and will be done in collaboration with the Immunology-Immunopathology-Immunotherapy (I3) lab (U959 Sorbonne Université-INSERM-APHP).
The Pr. Yves Allenbach will coordinate the projet.
Location
The Center of Research in Myology (CRM) and i3 laboratories are interested in muscle and systemic immune responses in auto-immune conditions. They developed approaches to identify pathological mechanisms, new biomarkers for diagnosis, and innovative targeted approaches.
Our team, headed by O. Benveniste and Y. Allenbach in CRM, is a world leader in cardiac and skeletal muscle toxicity of anti-tumor immunotherapy. It is a transversal research team, pioneer in the description and the management of ICI-induced myotoxicity (Allenbach Y et al. Nature Rev Immunol 2019; Salem JE et al. NEJM 2020). Our team have established one of the world's largest biobanks of patient tissues with myotoxicity.
I3 is lab expert in immunology with a unique expertise in identifying antigen-specific T cells through the analysis of T cell receptor repertoire using deep sequencing (Barennes P et al., Nature Biotechnology, 2020).
Both laboratories offer a unique interdisciplinary environment, with biologists, immunologists, clinicians, and bioinformaticians. The candidate will be based in the the Pitié‐Salpêtrière hospital campus in Paris which offers a dynamic environment with state-of-the-art facilities and platforms.
Project summary and position assignment
ICI-myotoxicity, combining skeletal and or cardiac inflammation, is a rare (0.5%) and unpredictable immune-related adverse events (irAE) but the most severe (up to 60% of fatality). Thus, it is crucial to identify predictive biomarkers and clarify the pathomechanisms. For this, our team and i3 lab have obtained jointly a considerable grant from the ARC Foundation for cancer research.
The project aims at :
- identifying specific pathways in cardiac and skeletal muscle tissue for rationale therapeutic targeting
- identifying predictive T cell clonotypes associated with ICI-myotoxicity
The successful candidate will play a pivotal role in a project focused on understanding the physiopathological mechanisms of immune-mediated muscular and cardiac complications of anti-cancer immunotherapy (Immune Checkpoint Inhibitors).
They will contribute to wet and dry lab-based research, including sample management, cell sorting, single cell, single nuclei, AIRRseq, RNASeq, as well as using available workflows developed by the host laboratory (PBMC and tissue 10X Genomics®) and implementing new strategies (Xenium, 10X Genomics®).
